氙灯发出一束光,通过光调制器和UV镜至滤光轮;滤过的光束进入光导纤维,向上至测量装置的透镜;光束从蜂窝板培养孔的底部向上透过样品悬浮液到达检测器,检测器得出浊度值后贮存于软件系统中供用户需要时调用。

测量装置沿着Y轴移动,同时载有蜂窝板的培养箱盘沿着X轴移动,这样就能在极短的时间内快速测量蜂窝板中的所有培养孔的浊度值。

1、能够排除培养基颜色变化对OD值的干扰

其他仪器通常在600 nm处测量菌悬液的浊度,但是600 nm处测量的不仅是浊度,而且某种程度上也包括培养液的颜色。Bioscreen采用宽波段滤光片技术专用于微生物学的浊度研究,因为其结果不会受到生长培养基颜色变化的影响,结果只表明浊度的生成状况。

2、样品不会产生冷凝或蒸发现象

技术确保蜂窝板盖子上不产生液体冷凝现象,冷凝现象会使结果失真。盖子也排除了样品蒸发现象,因为蒸发现象也会使结果失真。

3、***控温使得结果可靠度高

温度范围为15.0~65.0℃(设置范围:比环境温度低9℃~高于环境温度30℃),温度变化精度可控制在±0.1℃。传统的平板阅读器不能保持这些允许变量,因此结果可靠度低。

4、光学测量的选择

用户能选择2个波长另加1个宽波段(不受培养孔中颜色变化的影响)用于浊度测量。

5、1次可得到200个结果

传统的阅读器每运行1次可阅读1个96孔板,但是全自动生长曲线分析仪每次可测得200个结果。

6、可同时检测3个不同波长的OD值变化

可同时检测3个不同波长的OD值变化,因此可在检测微生物生长曲线的同时,还可检测1种物质生成(或降解)的动态变化 曲线、1种物质降解(或生成)的动态变化曲线。因此,可对微生物生长和相关物质代谢进行同步检测。

测试举例

全自动生长曲线分析仪是一个动态测量样品浊度的光学机械仪器。要明白她是否能进行一个特殊的测试,就有必要知道测试是否涉及培养液在生长期间的浊度(或颜色)变化。所有微生物(细菌、真菌、酵母和藻类)在培养液中繁殖都产生浊度。试管测试中凡是发生浊度和颜色变化的,大多数都能在她的“微测试试管”中来做。

除了多用途研究工作,全自动生长曲线分析仪也用于日常工作,如样品中细菌数量的测量(食品质量控制、细菌尿等)和微生物学测试(维生素、抗生素等化合物)。在很多其他研究中,如在测量抗生素对微生物的体外效果方面尤为突出。例如:

让我们设想一下我们需要体外检查5种新抗生素对于10种不同细菌的效果。为了比较,5种旧抗生素也用于测试。对于每种抗生素,我们将用5种不同的浓度;实验设置5个温度处理和3个pH处理;并且所有实验都用3个平行样。这样,这种抗生素效果模型所需要的测试数为:

10种抗生素(5种新抗生素和5种用于比较的旧抗生素);

10种细菌(例如:菌株1、…、菌株10);

用手工做这些测试(每天***多做20个测试)要花费22500÷20=1125个工作日。假定每年有200个工作日,这样就要花费1125÷200=5.6年+资料处理时间。如果用Bioscreen C做生长曲线耗时少于12h(如果采用连续振荡功能,这常能做到),每天就能做400个测试。Bioscreen C在56天内可做22500个这样的测试。这个实验用传统的手工方法要耗时6年,而用Bioscreen C可在2个月内完成。
